Formation of a new wound-healing hydrogel: biochemical interactions within a poly(acrylic acid) gel containing Adansonia digitata plant extract

Abstract
Background: Natural molecules and plants extracts have been loaded into biomaterials hydrogels depending on the characteristics of the natural molecules.

Objective: We propose to explain PAA gel containing Adansonia digitata plant extract spontaneous dissolution and the contribution of CaCO3 or KOH to gelation.

Methods: Dissolved PAA3000 at 2.5% form a gel on which 2.5 g of extract was added. Either 10mL of CaCO3 (100 mM) or 100 uL of KOH (1M) were added till the gelation. TLC analysis were done on plant extract. Acidity was determined and IR-ATR analysis were performed on gel containing or not plant extract.

Results: TLC identified phytomolecules belong to flavonoids, tanins, coumarins and terpens groups. IR-ATR spectra showed that CaCO3 obtained hydrogel, displayed more intensive –OH alcohol thanks to polyphenols addition and confirmed by aromatic C=C bands. Vaterite vibrational band became more intensive. For KOH obtained hydrogels, any characteristic vibrational band of alcohol–OH, acidic –OH nor ester C-O was present. Phenolic –OH and C=C groups characteristic vibrational bands appeared. Acidity decreased with extract addition.

Conclusion: The ideal hydrogel (pH=6.3) was obtained with KOH. In perspectives, we will study medical applications in wound healing process.
